چکیده

The scientific review presents the mechanisms of action cytoplasmic miRNAs, namely miRNA-mediated posttranslational silencing. To write article, information was searched using Scopus, Web Science, MedLine, PubMed, Google Scholar, EMBASE, Global Health, Cochrane Library, CyberLeninka databases. Translational inhibition by deadenylation-independent mechanisms is central to microRNA-mediated silencing in zebrafish.

MicroRNA (miRNA) is a class of small noncoding RNA approximately 22 nt in length. Animal miRNA silences complementary mRNAs via translational inhibition, deadenylation, and mRNA degradation. However, the underlying molecular mechanisms remain unclear. A key question is whether these three outputs are independently induced by miRNA through distinct mechanisms or sequentially induced within a sin...

MicroRNAs: Critical Regulators of mRNA Traffic and Translational Control with Promising Biotech and Therapeutic Applications

Context:MicroRNAs (miRNAs) are a class of short, endogenously-initiated, non-coding RNAs that post-transcriptionally control gene expression via translational repression or mRNA turnover. MiRNAs have attracted much attention in recent years as they play critical roles in gene expression and are promising tools with many biotech and therapeutic applications. The molecular mechanisms und...

Regulation of microRNA-mediated gene silencing by microRNA precursors

miRNAs are a class of about 1,000 noncoding RNAs that in mammals regulate about half of the protein-encoding genes by enhancing their degradation or preventing their translation1,2. After transcription, the long primary miRNA (pri-miRNA) transcripts are processed to 60to 80-nt precursor miRNAs (pre-miRNAs) and are subsequently cleaved to mature ~22-nt-long duplex RNAs3,4. With a few exceptions5...

MicroRNAs distinguish translational from transcriptional silencing during endotoxin tolerance.

We reported that gene-selective formation of facultative heterochromatin silences transcription of acute inflammatory genes during endotoxin (LPS) tolerance, according to function. We discovered that reversal of the epigenetically silenced transcription restored mRNA levels but not protein synthesis. Here, we find that translation repression of tumor necrosis factor-alpha (TNFalpha) occurs inde...

eIF4GI Facilitates the MicroRNA-Mediated Gene Silencing

MicroRNAs (miRNAs) are small noncoding RNAs that mediate post-transcriptional gene silencing by binding to complementary target mRNAs and recruiting the miRNA-containing ribonucleoprotein complexes to the mRNAs. However, the molecular basis of this silencing is unclear. Here, we show that human Ago2 associates with the cap-binding protein complex and this association is mediated by human eIF4GI...
